Background
This gene encodes a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. This membrane-bound protein has a neuregulin binding domain but lacks an active kinase domain. It binds ligands but does not transmit signals through phosphorylation. Instead, it forms heterodimers with other EGFR family members that possess kinase activity, leading to activation of pathways involved in cell proliferation and differentiation. Gene amplification or protein overexpression has been reported in various cancers, including prostate, bladder, and breast tumors. Alternative splicing produces different isoforms, one of which is secreted and modulates receptor activity.
