배경 정보

  • Domain: PTB domain mediates receptor interaction.
  • Function: DOK proteins act as adaptor/scaffolding proteins for assembling signaling complexes. DOK3 negatively regulates JNK signaling in B-cells via interaction with INPP5D/SHIP1 and may modulate ABL function.
  • Post-translational Modifications (PTM): Constitutively tyrosine-phosphorylated; upon IL2 stimulation, phosphorylated on C-terminal tyrosine residues possibly by Src kinases; can also be phosphorylated by ABL kinase.
  • Similarity: Belongs to DOK family, Type A subfamily; contains 1 IRS-type PTB domain and 1 PH domain.
  • Subunit Interaction: Upon tyrosine phosphorylation, interacts with CSK and INPP5D/SHIP1 via SH2 domains. Tyr-381 and Tyr-398 are required for INPP5D interaction, while only Tyr-381 is required for CSK interaction. Binds ABL through PTB domain in a kinase-dependent manner. Does not interact with RasGAP.
  • Tissue Specificity: Expressed in spleen.
