Target Information
Janus Activating Kinase 2 and 3 (JAK2/JAK3) are tyrosine kinases involved in cytoplasmic signal transduction. In response to cytokines or related factors (e.g., interferon, interleukins), JAKs are activated via phosphorylation at adjacent tyrosine residues. Activated JAKs phosphorylate STAT (signal transducers and activators of transcription) proteins, which dimerize and translocate to the nucleus to regulate gene transcription, including interferon-stimulated genes.
JAK2 is essential for IFN gamma-receptor complex initiation, while JAK1 acts as an amplifier. Active JAK1 may be required for complex responses, and some studies suggest that Tyk2 and JAK3 may substitute JAK2 under specific configurations in the receptor complex.
