Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 1 mM EDTA buffer (pH 8.0) for 10 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature. A suggested positive control is angiosarcoma or placenta tissue.
Target Information
VEGF (vascular endothelial growth factor) is a 45 kDa homodimeric, disulfide-linked glycoprotein involved in angiogenesis that promotes tumor progression and metastasis. VEGF influences vascular endothelium by promoting endothelial cell viability, mitogenesis, chemotaxis, and vascular permeability.
The VEGF family includes VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, and PIGF. VEGF and its receptor system (FLT1/VEGFR-1 and FLK1/KDR/VEGFR-2) are fundamental regulators of angiogenesis and are expressed mainly on endothelial cells.
Five isoforms of VEGF are generated by alternative splicing from a single VEGF gene. An alternately spliced form of FLT1 produces a soluble protein (sFLT1) that binds VEGF with high affinity, inhibiting angiogenesis.
Elevated VEGF levels are linked to POEMS (Crow-Fukase) syndrome, which affects multiple organs.
