Target Information

L-fucose is a key sugar in glycoproteins and other complex carbohydrates, involved in many functional roles such as cell-cell recognition. The fucosyl donor for these fucosylated oligosaccharides is GDP-β-L-fucose.
There are two pathways for GDP-fucose biosynthesis:

  • The major pathway converts GDP-α-D-mannose to GDP-β-L-fucose.
  • The alternate salvage pathway, present in certain mammalian tissues (e.g., liver, kidney), reutilizes L-fucose from glycoprotein and glycolipid turnover.
    This pathway involves phosphorylation of L-fucose to β-L-fucose-1-phosphate, followed by condensation with GTP by fucose-1-phosphate guanylyltransferase to form GDP-β-L-fucose.
