FEN-1 (Flap Endonuclease and Five prime Exonuclease)은 42 kDa 단백질로, 단일 가닥 5' 플랩을 포함하는 분기된 DNA 구조를 절단하는 엔도뉴클레이스 기능과 DNA 닉 및 갭 부위에서 작용하는 엑소뉴클레이스 기능을 수행합니다.
FEN-1의 엔도뉴클레이스 및 엑소뉴클레이스 활성은 핵 내의 PCNA(proliferating cell nuclear antigen)와의 결합에 의해 자극됩니다.
이 단백질은 DNA 복제 및 수선에 중요한 역할을 하며, 세포 자유 DNA 복제 시스템에서 필수적입니다.
