Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and plays a vital role in brain development.
Mutations, rearrangements, and amplifications in the ALK gene are associated with several tumors including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangement commonly produces a fusion gene between ALK (chromosome 2) and nucleophosmin (NPM, chromosome 5). The resulting NPM-ALK fusion protein is oncogenic.
ALK encodes a receptor tyrosine kinase with a transmembrane and extracellular domain, absent in the NPM-ALK fusion product. It shows the greatest sequence similarity to LTK and functions in neuronal development.
