Target Information
IRS-1 (Insulin Receptor Substrate 1) is a major substrate of the insulin receptor and is phosphorylated in response to insulin, IGF-1, and IL-4 stimulation. It undergoes phosphorylation on serine, threonine, and tyrosine residues across various tissues. Casein kinase II mediates part of the insulin-stimulated serine/threonine phosphorylation of IRS-1, with Thr502 and Ser99 identified as key phosphorylation sites. IRS-1 contains three potential 14-3-3 binding sites (Ser270, Ser374, Ser641), with the Ser270 site located in the phosphotyrosine-binding domain responsible for insulin receptor interaction.
