Product Specific Information

Recognizes wild type and mutant p53, cross-reacts with p53 in other primates. The epitope is within residues 18–30 of human p53.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation.
p53 up-regulates stress response genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization. p53 prevents proliferation of damaged cells, acting as an anticancer agent. Ser392 phosphorylation is associated with tumor formation.
Additionally, p53 is linked to aging, oxidative stress, and neural functions such as long-term potentiation (LTP).
It is expressed at low levels in normal cells but highly in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene lead to loss of DNA binding and tumor suppressor activity, frequently observed in human cancers and hereditary cancer syndromes such as Li-Fraumeni syndrome.
