Target Information
The MAP kinase-interacting kinases (MKNK1 and MKNK2) are activated by MAP kinases and regulate protein synthesis by phosphorylating eIF4E on Ser209, potentially contributing to tumorigenesis. Overexpression of Mnk2 increases phosphorylation of endogenous eIF4E, showing its role as an eIF4E kinase in vivo. Mnk2 may also be involved in responses to environmental stress and cytokines. Expression of active mutants of MNK1 and MNK2 reduces cap-dependent translation relative to cap-independent translation in transient reporter assays. Human Mnk2 shares approximately 94% identity with murine Mnk2 and 71% with human Mnk1. In vitro phosphorylation studies indicate that Mnk2 is a better substrate than Mnk1 for Erk2, p38MAPKα, and p38MAPKβ.
