Target Information
SIAH는 초파리의 seven in absentia (sina) 유전자의 인간 상동체로, 장 상피세포에서 발현되며 세포자멸사 과정에서 활성화되는 종양 억제 단백질입니다. 인간 SIAH 단백질은 SIAH-1과 SIAH-2의 두 가지 유전자 산물로 존재하며, C-말단 서열은 보존되어 있으나 N-말단 영역에서 차이를 보입니다.
SIAH-1은 32 kDa 단백질로, 단백질 분해에 필요한 N-말단 RING-finger 도메인과 SIAH 결합 및 올리고머화를 조절하는 C-말단 cysteine-rich 도메인을 포함합니다. 종양 억제 단백질로서 SIAH-1은 DCC(deleted in colorectal cancer) 단백질과 결합하여 유비퀴틴-프로테아좀 경로를 통해 DCC 분해를 조절합니다. 또한 Bcl-2 관련 단백질 Bag-1과 결합하여 세포 성장을 억제합니다. 대부분의 SIAH-1은 핵에 위치하지만 일부는 세포질에서도 검출됩니다. 이러한 핵 내 위치는 SIAH 단백질이 다른 핵 매트릭스 단백질 및 DNA와 상호작용할 가능성을 시사합니다.
