Target Information
Semaphorins are the largest family of axon guidance cues known. They are divided into eight classes, with classes 3–7 found in vertebrates.
- Class 3 Semaphorins are secreted proteins.
- Classes 4–6 are transmembrane proteins.
- Class 7 is membrane-associated via glycosylphosphatidylinositol (GPI) linkage.
Semaphorins share a conserved ~400 amino acid sema domain and are known as collapsing factors and mediators of axon repulsion, while also functioning as context-dependent chemoattractants. They play roles in cardiovascular development and immune cell antigen presentation. Their signaling is mediated by Neuropilin and Plexin receptor families.
