Target Information
Longevity assurance genes (LAGs) are a subgroup of the homeobox gene family. Five mammalian homologs have been identified, with key roles in ceramide synthesis. LAG family proteins are also implicated in cell growth regulation and cancer differentiation.
LASS6 may be involved in sphingolipid synthesis or its regulation. It interacts with ATP6V0C, ASGR1, ASGR2, and SLC22A1/OCT1, and localizes to the nuclear membrane as a multi-pass membrane protein, possibly expressed on the endoplasmic reticulum membrane. Glycosylation on Asn-18 is not essential for function.
Like other LAG family members, LASS6 contains a homeodomain and LAG1 domain and shows highest homology to LASS5. The gene is localized to human chromosome 2q24.3, spanning approximately 318 kb. Sequence conservation in mouse and zebrafish suggests evolutionary conservation across vertebrates. LASS6 is broadly expressed in various tissues and is associated with cancer differentiation and early embryonic development.
