For FACS analysis, use 10 µL of the suggested working dilution to label 1×10^6 cells in 100 µL.
Rat anti-Mouse CD40 antibody, clone 3/23, recognizes the murine CD40 cell surface glycoprotein.

Target Information

The CD40 antigen is a single-chain glycoprotein belonging to the tumor necrosis factor/nerve growth factor superfamily. It shows significant homology to the Hodgkin’s disease-associated antigen CD30.
CD40 is expressed on all B cells except plasma cells, and also on some epithelial cells, carcinomas, and lymphoid dendritic cells. It plays essential roles in immune and inflammatory responses, including:

• T cell-dependent immunoglobulin class switching
• Memory B cell development
• Germinal center formation

AT-hook transcription factor AKNA regulates CD40 expression, contributing to homotypic cell interactions. Adaptor protein TNFR2 interacts with CD40 to mediate signal transduction.
The CD40–CD40L interaction is necessary for amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
