Target Information

Nucleophosmin (NPM1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, nucleocytoplasmic trafficking, regulation of DNA polymerase alpha activity, centrosome duplication, and p53 regulation.
NPM1 shuttles between the nucleus, cytoplasm, and nucleolus, acting as a chaperone for core histones. It binds nucleic acids, prevents protein aggregation, protects enzymes during thermal stress, and assists in protein renaturation.

NPM1 is associated with the centrosome and serves as a substrate for CDK2/cyclin E during centrosome duplication. It participates in tumor-associated chromosomal translocations forming fusion proteins such as NPM-ALK, NPM-RAR, and NPM-MLF1, contributing to oncogenic processes.
Additionally, NPM1 sequesters tumor suppressor RF in the nucleolus, protecting it from degradation. Dysfunction of NPM1 is linked to diseases including acute myeloid leukemia and lymphomatoid papulosis.
