Product Specific Information

The monoclonal antibody MH5A reacts with a homolog of PD-1 (PD-1H), also known as VISTA. PD-1H is a 309 aa type I transmembrane protein belonging to the CD28 superfamily. It is expressed on nearly all hematopoietic cells except B cells and negatively regulates T cell responses by suppressing early TCR activation, inhibiting proliferation, and reducing TH1 cytokine production.

MH5A antibody has been shown to inhibit T cell infiltration in graft-versus-host disease mouse models.

  • Use ≤ 1 µg/test (100 µL staining volume, 10⁵–10⁸ cells/test)

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). It can substitute for PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Fixation:

Store up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency. Clone-specific performance should be confirmed empirically.

Target Information

VISTA (V-domain Ig containing suppressor of T-cell activation), also known as B7-H5 or PD-1H, is encoded by the VSIR gene. Although homologous to PD-L1, VISTA has a distinct expression pattern. In both mouse and human, it is predominantly expressed on hematopoietic cells—highest on myeloid cells, lower on T cells, and absent on B cells.

VISTA on antigen-presenting cells directly suppresses CD4⁺ and CD8⁺ T cell proliferation and cytokine production, making it a promising target for cancer immunotherapy. Known ligands include CD28H, VSIG-3, and VSIG-8.
