Invitrogen™ Low Density Lipoprotein from Human Plasma, BODIPY™ FL complex (BODIPY™ FL LDL)

Human low-density lipoprotein (LDL) is a large protein complex (∼500,000 Da) that binds to specific receptors on vertebrate cell surfaces and delivers cholesterol via receptor-mediated endocytosis.
Labeled LDL complexes are useful tools for studying this phenomenon.

Experimental Applications

  • Fluorescently labeled LDL can be added to cultured cells for analysis by microscopy or flow cytometry.
  • Alternatively, labeled LDL can be injected into test animals to analyze label distribution over time.

We offer unlabeled LDL and two classes of labeled LDLs:

  1. Unmodified apoprotein LDL – used to study normal cholesterol delivery and internalization.
  2. Acetylated (Ac) LDL – used to study cell types expressing receptors specific for acetylated LDL (e.g., endothelial and microglial cells).

Fresh LDL Production

LDL and AcLDL products are prepared from fresh human plasma approximately every two months.
Nonacetylated LDL products are shipped within two weeks of preparation, while acetylated LDL products are available continuously.

Nonacetylated vs. Acetylated LDL

  • Nonacetylated LDL: Used to study normal cholesterol delivery and internalization.
  • Acetylated LDL: Lysine residues of LDL's apoprotein are acetylated, preventing binding to the LDL receptor. Instead, uptake occurs via scavenger receptors on endothelial and microglial cells.

Key Applications for Labeled LDL

  • Counting and tracking cell-surface LDL receptors and their clustering after internalization
  • Quantitating LDL receptor activity in fibroblasts (alternative to radiolabeled LDL assay)
  • Investigating LDL expression and identifying receptor deficiencies in various cell lines
