Product Specific Information

Description:
The 9E2 monoclonal antibody reacts with human CD335 (NKp46), a member of the natural cytotoxicity receptor family (including NKp30 and NKp44). CD335 is expressed on NK cells and plays a role in activating and expanding resting NK cells upon interaction with dendritic cells, enabling elimination of tumor and virus-infected cells (e.g., influenza). It also interacts with pancreatic beta cells during type I diabetes development.

The 9E2 antibody has been reported to induce cytotoxicity.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use 5 µL (0.25 µg) per test (defined as the amount that stains a cell sample in 100 µL final volume).
Cell number should be determined empirically (10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 436 can be excited with the violet laser line (405 nm) and emits at 436 nm.
Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

NKp46 (CD335, NCR1) is a cytotoxicity-activating receptor that enhances the efficiency of activated NK cells in tumor cell lysis. It consists of two Ig-like domains related to leukocyte immunoglobulin-like (LIR) and killer inhibitory receptors (KIR).
