Target Information
IFI16 (gamma-interferon-inducible protein 16) binds double-stranded DNA, preferentially to supercoiled and cruciform DNA structures. It is implicated in transcriptional regulation and repression, hematopoietic differentiation, and cell cycle control through modulation of p53/TP53 and retinoblastoma protein functions.
IFI16 also plays a role in innate immune response by recognizing viral dsDNA in the cytosol and nucleus, recruiting TMEM173/STING, and mediating IFN-beta induction. It is essential for activation of the IFR3 signaling cascade during HSV-1 infection, promoting heterochromatin assembly on viral DNA and inhibiting viral gene expression and replication.
Additionally, IFI16 is involved in MTA1-mediated epigenetic regulation of ESR1 expression in breast cancer.
