Target Information
IFI16 (gamma-interferon-inducible protein 16) binds double-stranded DNA, preferentially to supercoiled and cruciform DNA structures.
It is involved in transcriptional regulation and acts as a transcriptional repressor. IFI16 may regulate hematopoietic differentiation by activating unknown target genes and controls cellular proliferation through modulation of p53/TP53 and retinoblastoma protein functions.
IFI16 also participates in innate immune responses by recognizing viral dsDNA in the cytosol and nucleus. Upon binding viral DNA, it recruits TMEM173/STING and induces IFN-beta expression.
Additionally, IFI16 activates the IFR3 signaling cascade during HSV-1 infection, promotes heterochromatin assembly on herpesviral DNA, and inhibits viral immediate-early gene expression and replication.
IFI16 is further implicated in MTA1-mediated epigenetic regulation of ESR1 expression in breast cancer.
