Target Information
TAB2 is an activator of MAP3K7/TAK1, required for IL-1-induced activation of NF-kappaB and MAPK8/JNK. It forms a kinase complex with TRAF6, MAP3K7, and TAB1, serving as an adaptor linking MAP3K7 and TRAF6. TAB1 and MAP3K7 also participate in TNFSF11/RANKL-induced signaling through activation of TNFRSF11A/RANK, influencing osteoclast development and function.
Recent studies show TAB2 and TAB3 interact with Beclin-1, an autophagy mediator. During autophagy induction, TAB2/TAB3 dissociate from Beclin-1 and bind TAK1. Overexpression inhibits autophagy, while depletion triggers it, suggesting TAB2/TAB3 act as regulatory points for autophagy.
