Target Information
TAB2 is an activator of MAP3K7/TAK1, required for IL‑1–induced activation of NF‑κB and MAPK8/JNK. It forms a kinase complex with TRAF6, MAP3K7, and TAB1, functioning as an adaptor linking MAP3K7 and TRAF6. TAB2, TAB1, and MAP3K7 also participate in TNFSF11/RANKL‑induced signaling via TNFRSF11A/RANK, influencing osteoclast development and function.
Recent studies show TAB2 and TAB3 interact with Beclin‑1, an autophagy mediator. Upon autophagy induction, they dissociate from Beclin‑1 and bind TAK1. Overexpression of TAB2/TAB3 inhibits autophagy, while depletion triggers it, suggesting a regulatory role in autophagy control.
