IRS-1는 인슐린 수용체의 주요 기질로, 인슐린, IGF-1 및 IL-4 자극에 의해 인산화됩니다. 다양한 조직에서 세린, 트레오닌, 티로신 잔기에서 인산화가 일어납니다. Casein kinase II는 IRS-1의 일부 인슐린 자극 세린/트레오닌 인산화를 매개하며, Thr 502와 Ser 99가 주요 인산화 부위로 확인되었습니다. IRS-1은 14-3-3 단백질 결합 부위를 세 곳(Ser 270, Ser 374, Ser 641)에 가지며, Ser 270 주변 모티프는 인슐린 수용체와 상호작용하는 인산티로신 결합 도메인에 위치합니다.
