Target Information
Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases that serve as potential drug targets for allergic disorders and autoimmune diseases. SYK may also play a role in cancer.
Upon activation of B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI), Syk translocates to the plasma membrane and phosphorylates downstream adaptor proteins, initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, and gene transcription.
Syk interacts with receptor assemblies through its SH2 domains binding to ITAM motifs phosphorylated by Src-family kinases, which also activate Syk via phosphorylation of its activation loop.
