Target Information

ALAS-1 is a nuclear-encoded mitochondrial matrix enzyme that catalyzes the condensation of glycine with succinyl-CoA to form delta-aminolevulinate, CO₂, and CoA. It regulates the first and rate-limiting step of the heme biosynthetic pathway.
This enzyme is one of two ALAS isoforms and functions as a pyridoxal-5'-phosphate–dependent housekeeping enzyme. It is ubiquitously expressed and provides heme for cytochromes and other hemoproteins.
ALAS1 in the liver is negatively regulated by heme. Transcription of the ALAS1 gene is stimulated by cAMP and respiratory uncoupling, while phorbol esters and insulin repress its expression.
ALAS1 is implicated in the reciprocal regulation of heme biosynthesis and the circadian clock, suggesting potential relevance for circadian disorder treatments. Downregulation of ALAS1 expression has been observed in acute liver failure, affecting heme metabolism and liver function.
