Target Information
Surface Layer Proteins (S-layers) are two-dimensional arrays of protein or glycoprotein subunits forming symmetric, porous, lattice-like layers that completely cover the cell surface of prokaryotes. These subunits are held together and attached to cell wall carbohydrates by non-covalent interactions and can spontaneously reassemble in vitro through entropy-driven processes.
In Lactobacillus species, S-layer proteins vary in size and possess high predicted isoelectric points (pI). The positive charge is concentrated in the conserved cell wall binding domain, which may be located at either the N- or C-terminal depending on the species. The variable domain facilitates self-assembly into periodic structures.
Although the biological functions of Lactobacillus S-layer proteins are not completely understood, they may mediate bacterial adherence to host cells or extracellular matrix components and exhibit protective or enzymatic roles. These proteins show promise as antigen carriers in live oral vaccine design due to their adhesive and immunomodulatory properties and the non-pathogenic nature of Lactobacillus species.
