Product Specific Information

• This antibody does not react with rat.
• For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

HLA-DR, a major histocompatibility complex (MHC) class II molecule, is composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
The alpha chain gene contains five exons:

1. Exon 1 encodes the leader peptide.
2. Exons 2 and 3 encode extracellular domains.
3. Exon 4 encodes the transmembrane domain and cytoplasmic tail.

DRA lacks polymorphisms and serves as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5.
The beta chain carries polymorphisms that determine peptide binding specificity.
Hundreds of DRB1 alleles exist, and typing these polymorphisms is standard in bone marrow and kidney transplantation.
HLA-DR is expressed mainly on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
MHC class II molecules (DR, DQ, DP) bind intracellularly processed peptides, present them to T-helper cells, and play a critical role in initiating immune responses.
