- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min.
Target Information
Beta-catenin is an adherens junction (AJ) protein crucial for cell-cell adhesion and epithelial layer maintenance. It interacts with E-cadherin and alpha-catenin to anchor the adhesion complex to the actin cytoskeleton. Beta-catenin also associates with signaling complexes involving APC and microtubules, participating in Wnt/Wingless pathways that regulate morphogenesis and cell proliferation.
Beta-catenin expression is observed in various hair follicle cells and epithelial structures. Abnormal beta-catenin activity has been linked to pilomatrixoma, medulloblastoma, colorectal, ovarian, and other solid tumors. In the nucleus, beta-catenin coactivates Lef/Tcf transcription factors, promoting transcription of genes such as cyclin D and c-myc, contributing to tumorigenesis.
