Target Information
The CDKN2A gene produces multiple transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins. Two isoforms act as inhibitors of CDK4 kinase, while another variant encodes an alternate open reading frame (ARF) product that stabilizes the tumor suppressor protein p53 by interacting with and sequestering the E3 ubiquitin-protein ligase MDM2. Despite structural and functional differences, both CDK inhibitor isoforms and ARF product contribute to G1 phase cell cycle control. Mutations or deletions in this gene are common in various tumors, underscoring its role as an important tumor suppressor.
