Product Specific Information

  • Membrane permeabilization is required for flow cytometry applications.
  • For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
  • Mouse anti-Human Glucocorticoid Receptor antibody, clone 5E4, recognizes the human glucocorticoid receptor (NR3C1), a 777 amino acid receptor with three distinct functional domains:
    • N-terminal modulating domain
    • DNA-binding domain
    • C-terminal steroid-binding domain

Target Information

Glucocorticoid Receptor (NR3C1) is a receptor for glucocorticoids that functions as both a transcription factor and a regulator of other transcription factors. It is typically found in the cytoplasm and translocates to the nucleus upon ligand binding. The receptor is expressed in various cardiac tissues including atria, ventricles, aorta, and septum.
Alternative splicing and promoter usage generate multiple transcript variants. Mutations in the NR3C1 gene are associated with glucocorticoid resistance. The receptor forms complexes with chaperone proteins and binds steroids such as dexamethasone with nanomolar affinity.
