Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon activation, p53 induces transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

p53 consists of an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain. Phosphorylation at Ser15 is critical for its activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation.

p53 functions to block proliferation of damaged cells, acting as an anticancer agent. It is expressed at low levels in normal cells but highly upregulated in transformed or malignant cells. Mutations in the TP53 gene often lead to loss of tumor suppressor activity and are common in human cancers. Germline TP53 mutations are associated with cancer-prone syndromes such as Li-Fraumeni syndrome.
