Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the αLβ2 integrin. CD11a is implicated in activation of the LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAMs 1–3. It is also involved in lymphocyte costimulatory signaling, cell differentiation, proliferation, and T-cell effector functions.
Blocking CD11a function has become an important therapeutic approach in multiple inflammatory diseases. For example, the humanized anti-CD11a antibody Efalizumab (Raptiva) interferes with T-cell migration to inflammation sites, while simvastatin binding to CD11a leads to immunomodulation.
Two transcript variants encoding different isoforms of CD11a have been identified. Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
