Target Information
ALAS1 (Aminolevulinate Synthase 1) is a nuclear-encoded mitochondrial matrix enzyme that catalyzes the condensation of glycine with succinyl-CoA to form δ-aminolevulinate, CO₂, and CoA.
It regulates the first and rate-limiting step of the heme biosynthetic pathway. As one of two ALAS isoforms, ALAS1 is a pyridoxal-5'-phosphate-dependent housekeeping enzyme expressed ubiquitously, providing heme for cytochromes and other hemoproteins.
ALAS1 in liver is negatively regulated by heme and transcriptionally stimulated by cAMP and respiratory uncoupling. Phorbol esters and insulin repress ALAS1 gene expression.
ALAS1 may be involved in reciprocal regulation of heme biosynthesis and the circadian clock, suggesting a potential therapeutic target for circadian disorders.
Downregulation of ALAS1 expression in acute liver failure is associated with altered heme metabolism and impaired liver function.
