Target Information
The human AF9 gene is one of the most common fusion partner genes with the ALL1 (MLL) gene at 11q23, resulting in t(9;11)(p22;q23).
AF9 spans over 100 kb and contains two patient breakpoint cluster regions (BCRs):
- BCR1: within intron 4 (site A)
- BCR2: spans introns 7 and 8 (site B)
Structural features include:
- In vivo DNA topo II cleavage site and in vitro DNase I hypersensitive site in intron 7 (BCR2)
- Scaffold associated regions (SARs): SAR1 in intron 4 and SAR2 encompassing exons 5–7
These regions share structural similarities with MLL BCR, supporting a DNA breakage and repair model for nonhomologous recombination between MLL and AF9.
