Description:
The eBioH4B4 monoclonal antibody reacts with human CD107b (LAMP-2), a highly glycosylated type I transmembrane protein (~105 kDa) located in lysosomal/endosomal membranes. It is expressed in nearly all cells and on the surface of degranulating T cells, activated platelets, and some cancer cells. Mutations in CD107b cause Danon disease, a lysosomal glycogen storage disorder.
Applications Reported:
This antibody is suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-titrated and validated for intracellular staining and flow cytometric analysis of Jurkat cells. Recommended use: 5 µL (0.125 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.
Fluorophore Information:
eFluor® 660 replaces Alexa Fluor® 647. It emits at 659 nm and is excited by the red laser (633 nm).

Target Information
LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b):
A member of the lysosomal membrane glycoprotein family, LAMP2 plays roles in lysosomal protection, maintenance, and adhesion. It provides selectins with carbohydrate ligands and may contribute to tumor metastasis. Mature LAMP2 (100–130 kDa) is heavily glycosylated and consists of:
- A large amino-terminal intra-lysosomal domain
- A hydrophobic transmembrane domain
- A short cytoplasmic tail
LAMP2 is critical for chaperone-mediated autophagy and lysosomal-phagosomal fusion. Deficiency leads to autophagosome accumulation and Danon’s disease (cardiomyopathy and myopathy). LAMP2 has three isoforms: LAMP-2A, LAMP-2B, and LAMP-2C.
