Background
Mitogen-activated protein kinase (MAPK) signaling cascades include MAPK (ERK), MAPK kinase (MKK or MEK), and MAPK kinase kinase (MAPKKK or MEKK). MAPKKK5 phosphorylates and activates its downstream kinase (MKK4), which subsequently activates c-Jun N-terminal kinase (JNK)/stress-activated protein kinase (SAPK). MAPKKK5 does not activate MAPK/ERK.
MAPKKK5 consists of 1,374 amino acids containing all 11 kinase subdomains and is highly conserved across species. Northern blot analysis shows abundant MAPKKK5 transcript expression in human heart and pancreas.
