Localized to protruding lamellipodia and filopodia tips, neuronal growth cones, and synaptosomes. May shuttle between postsynaptic densities and nucleus (By similarity).
Background
This gene encodes a member of the Abelson-interactor family of adaptor proteins. These proteins facilitate signal transduction as components of multiprotein complexes and regulate actin polymerization and cytoskeletal remodeling through interactions with Abelson tyrosine kinases.
The encoded protein plays a role in macropinocytosis as part of the WAVE2 complex and forms a complex with EPS8 and SOS1 mediating Ras-to-Rac signal transduction.
Abi-1 may be involved in progression of malignancies such as melanoma, colon cancer, and breast cancer. A t(10;11) translocation involving this gene and MLL has been linked to acute myeloid leukemia.
Alternative splicing results in multiple isoforms, and a pseudogene is located on chromosome 14q.
