Cellular Localization Cytoplasm and nucleus. Cytoplasmic and nuclear in the absence of TGF-β. Upon TGF-β stimulation, migrates to the nucleus when complexed with SMAD4. Released from the SMAD2/SMAD4 complex via PPM1A and exported from the nucleus through RANBP1 interaction. Co-localizes with LEMD3 at the nuclear inner membrane. MAPK-mediated phosphorylation has no effect on nuclear import. PDPK1 prevents nuclear translocation in response to TGF-β. Mainly localized to the nucleus in early embryo development, later evident in cytoplasm of the inner cell mass at the blastocyst stage.
Background The SMAD3 protein belongs to the SMAD family, homologous to Drosophila mothers against decapentaplegic (Mad) and C. elegans Sma. SMAD proteins are signal transducers and transcriptional modulators mediating multiple signaling pathways. SMAD3 acts as a transcriptional modulator activated by TGF-β and is implicated in carcinogenesis regulation. (RefSeq, Apr 2009)
