Background

  • Function: Required for DNA recombination, repair, and replication. RP-A activity is mediated by single-stranded DNA binding and protein interactions.
  • Post-translational Modification: Phosphorylated in a cell cycle-dependent manner (S phase to mitosis). ATR phosphorylates the protein upon DNA damage, promoting translocation to nuclear foci. Can also be phosphorylated in vitro by PRKDC/DNA-PK (with Ku and DNA) and by CDC2.
  • Subcellular Location: Present in PML nuclear bodies; redistributes to discrete nuclear foci upon DNA damage.
  • Subunit Structure: Heterotrimer of 70, 32, and 14 kDa chains. DNA-binding activity may reside mainly in the 70 kDa subunit. Interacts with SERTAD3/RBT1 and TIPIN.
