세포 내 위치

Cytoplasm, Nucleus. Shuttles between the nucleus and cytoplasm. Translocated into the nucleus upon tyrosine phosphorylation and dimerization in response to activated FGFR1–4 signaling. Constitutive nuclear presence is independent of tyrosine phosphorylation. Predominantly cytoplasmic without stimuli. Upon LIF stimulation, accumulates in the nucleus. The BART–ARL2 complex is important for nuclear translocation and retention of STAT3. Identified in a complex with LYN and PAG1.

배경 (Background)

The STAT3 protein is a member of the STAT family, activated by phosphorylation in response to cytokines and growth factors such as IFNs, EGF, IL5, IL6, HGF, LIF, and BMP2. Once activated, STAT3 forms dimers that translocate to the nucleus and act as transcription activators. It regulates expression of genes involved in cell growth and apoptosis. Rac1 binds and regulates STAT3 activity, while PIAS3 acts as a specific inhibitor. Mutations in STAT3 are associated with infantile-onset multisystem autoimmune disease and hyper-IgE syndrome.
