Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor (~90 kDa) has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. It is a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

The AR signaling pathway is essential for male reproductive organ development and function, including the prostate and epididymis, and also affects nonreproductive tissues such as muscle, hair follicles, and brain. AR is a phosphoprotein and regulates MAP kinase activity. Inhibition of the MEK1/2 pathway alters AR phosphorylation. Dysregulation of AR signaling is linked to diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations can cause complete androgen insensitivity syndrome (CAIS).
