The R-phycoerythrin (PE)-Texas Red conjugate permits simultaneous multicolor labeling and detection of multiple targets with excitation by a single excitation source — the 488 nm line of the argon-ion laser.


Target Information

HLA-DR, a major histocompatibility complex (MHC) class II molecule, is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).

  • The alpha chain gene contains 5 exons:
    • Exon 1 encodes the leader peptide
    • Exons 2 and 3 encode the extracellular domains
    • Exon 4 encodes the transmembrane domain and cytoplasmic tail
  • DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5, and lacks polymorphisms in the peptide-binding region.
  • The beta chain contains all the polymorphisms determining peptide-binding specificity. Hundreds of DRB1 alleles have been described, and typing is routinely performed for bone marrow and kidney transplantation.
  • HLA-DR is primarily expressed on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
  • The human class II region encodes DR, DQ, and DP loci, which bind intracellularly processed peptides and present them to T-helper cells, initiating immune responses.

HLA and MHC antibodies are crucial in Immunopeptidomics, aiding in the identification and characterization of neoantigens through high-performance liquid chromatography coupled with tandem mass spectrometry.
