Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, participating in signal regulation and receptor removal from the cell surface.
CD33 is expressed on cells of the myelomonocytic lineage and binds sialic acid residues in N- and O-glycans. It is a therapeutic target for acute myeloid leukemia (AML) and is present on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells.
CD33 serves as a marker for peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
