Target Information
Ago 단백질은 체세포에서 널리 발현되며, miRNA와 결합하여 다양한 RNA 침묵화 경로에 관여합니다. 그중 Ago2 단백질만이 endonucleolytic 또는 “Slicer” 활성을 가지며, 완전한 상보성을 가진 miRNA에 의해 표적 mRNA를 절단할 수 있습니다. 부분적 상보성의 경우 절단 대신 번역 억제 기능을 수행합니다.
Ago2는 RNA 간섭, 번역 억제, 이질염색질화 등 소형 RNA 기반 유전자 침묵화 과정에서 핵심적인 역할을 하며, RISC 복합체의 주요 구성요소로서 siRNA 또는 miRNA에 의해 유도된 mRNA 분해를 촉진합니다. 또한 Ago2는 Dicer 비의존적 RNA 절단 효소로 작용하며, miRNA 성숙 조절에도 관여합니다.
Ago2의 과발현은 암세포 성장 및 환자의 생존율과 관련이 있으며, 마우스 모델에서의 유전자 결손 연구는 Ago2가 배아 발달 및 B-림프구, 적혈구 발달의 핵심 조절자임을 보여줍니다.
