Target Information
MAP4K3 (also known as GLK) is a member of the STE20 family of serine/threonine protein kinases containing an N-terminal catalytic domain and a C-terminal regulatory domain. It acts upstream of MEKK1 and is stimulated by UV radiation and TNF-alpha, leading to activation of the JNK signaling pathway.
MAP4K3 can directly activate PKC-T during TCR signaling. Enhanced MAP4K3 expression and activation of PKC-T and IKK have been observed in T cells from individuals with systemic lupus erythematosus.
MAP4K3 functions as a pro-apoptotic kinase that activates BAX through modulation of PUMA, BAD, and BIM. Its kinase activity can be stimulated by UV radiation and TNF-alpha, specifically activating the c-Jun N-terminal kinase (JNK) pathway. It functions upstream of mitogen-activated protein kinase kinase 1 (MEKK1).
Previously known as RAB8-interacting protein-like 1 (RAB8IPL1), it has been renamed mitogen-activated protein kinase kinase kinase 3 (MAP4K3).
