Target Information

RNA polymerase (Pol) III synthesizes tRNA, 5S rRNA, 7SL RNA, and U6 snRNA, and is overexpressed in many transformed cell lines and tumors. To maintain rapid growth, cells require high levels of Pol III-transcribed RNA, dependent on TFIIIB and TFIIIC2 transcription factor complexes.
TFIIIC2 consists of five subunits (TFIIIC220, TFIIIC110, TFIIIC102, TFIIIC90, TFIIIC63), which are overexpressed in adenovirus-transformed and malignant cells such as ovarian carcinomas.
TFIIIC2 recruits RNA Pol III and TFIIIB to promoter elements, potentially contributing to malignant cell deregulation. TFIIIB includes TBP, BRF1 (TFIIIB90), and TFIIIB subunits, whose expression is upregulated in transformed cells.
In many carcinomas, tumor suppressors RB and p53 are inactivated, affecting their ability to bind and inhibit TFIIIB function.
