Target Information
The SMAD1 protein belongs to the SMAD family, which is homologous to the Drosophila gene mothers against decapentaplegic (Mad) and the C. elegans gene Sma. SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways.
SMAD1 mediates bone morphogenetic protein (BMP) signaling, which regulates cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP ligand binding, SMAD1 is phosphorylated by BMP receptor kinases and forms a complex with SMAD4 to regulate transcription.
SMAD1 is targeted by E3 ubiquitin ligases such as SMURF1 and SMURF2, leading to ubiquitination and proteasome-mediated degradation. Alternatively spliced transcript variants encoding the same protein have been observed.
