Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)는 Alzheimer’s disease 환자의 뇌에서 발견되는 아밀로이드 플라크의 주요 구성 성분입니다. Abeta는 40–43개의 아미노산으로 구성되며, beta-amyloid precursor protein (beta APP)으로부터 두 단계의 절단 과정을 통해 생성됩니다.
첫 번째 절단은 beta-secretase (BACE)에 의해 수행되며, 이는 propeptide 형태로 합성되어 furin에 의해 활성형으로 전환됩니다. 이후 gamma-secretase 복합체(주요 구성 요소: presenilin-1, presenilin-2, nicastrin 등)에 의해 세포막 내에서 두 번째 절단이 일어나 amyloid beta peptide가 생성됩니다.
Abeta는 세포 외 필라멘트성 단백질로, amyloid cores, neuritic plaques, neurofibrillary tangles 등에서 발견됩니다. Alzheimer’s disease 외에도 Lewy body dementia, Down’s syndrome, cerebral amyloid angiopathy 등에서도 관찰됩니다.
