Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR acts as a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

AR signaling is essential for development and function of male reproductive organs such as the prostate and epididymis, and also plays roles in muscle, hair follicles, and brain. AR is a phosphoprotein that regulates MAP kinase activity; inhibition of MEK1/2 affects AR phosphorylation. Abnormal AR signaling is linked to diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in AR gene are associated with complete androgen insensitivity syndrome (CAIS).
