Product Specific Information

PA1-978 detects proteasome 20S Y from purified bovine and human 26S proteasome samples.
It has been successfully used in Western blot and ICC/IF procedures.
By Western blot, this antibody detects an approximately 25 kDa protein representing proteasome 20S Y from purified human 26S proteasome.

The PA1-978 immunizing peptides correspond to amino acid residues 41–58 from human proteasome 20S Y and residues 74–91 from frog.
These sequences are completely conserved in mouse, rat, and zebrafish.
The PA1-978 immunizing peptide (Cat. # PEP-173) is available for use in neutralization and control procedures.


Target Information

Proteolytic degradation is essential for maintaining proper levels of short-lived and regulatory proteins involved in metabolism, stress response, antigen presentation, receptor modulation, cell cycle regulation, transcription, and signaling.
The ubiquitin-proteasome pathway degrades most cytosolic and nuclear proteins in eukaryotic cells, while other proteins are processed via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.

The 26S proteasome is an ATP-dependent, multisubunit (~31) molecular complex (~2.5 MDa) composed of a 20S proteolytic core capped by one or two 19S regulatory subunits.
The 19S complex recognizes ubiquitinated proteins and mediates their unfolding and translocation into the 20S core.
The PA28/11S REG Activator complex (α, β, γ subunits) enhances 20S proteolytic activity.

Defects in the ubiquitin-proteasome pathway are associated with several genetic diseases, including cystic fibrosis (CFTR), Angelman’s syndrome (E6-AP), and Liddle syndrome (ENaC).
